Intended uses

This HMGB1 ELISA kit is a 1.5-hour solid-phase ELISA designed for the quantitative determination of bovine HMGB1. This ELISA kit is for research use only, not for therapeutic applications!

Preparation and storage

Store all reagents at 2-8 degrees C.

Sample preparation

We suggest prior experimentation with clean (undiluted) samples, 1: 2 or 1: 4 dilutions. Avoid diluting your samples more than 1:10, as this would exceed the dilution limit established for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team.

Product note

Information selected from the online data sheet is extracted from bioinformatics databases, occasionally resulting in ambiguous or irrelevant product information. It is the customer’s responsibility to review, verify and evaluate the information to make sure it matches their requirements before purchasing the kit. The essays in our ELISA kit are dynamic research tools and can sometimes be updated and improved.

If the format of this assay is important to you, please request the current manual or contact our technical support team with a pre-sale consultation before placing an order. We will confirm the current details of the trial. We cannot guarantee that the sample manual posted online is the most current manual, it is intended to serve as an example only. See the instructions for use provided with the test kit for precise details.

Other notes

Occasionally, small volumes of the HMGB1 ELISA kit vials may become trapped in the product vial seal during shipping and storage. If necessary, briefly centrifuge the vial in a benchtop centrifuge to dislodge any liquid on the lid of the container. Some products may need to be shipped with dry ice and an additional fee may apply for dry ice.

MBS738320 is a ready-to-use microwell plate ELISA (enzyme-linked immunosorbent assay) kit to test for the presence of target analytes of the High Mobility Group Box1 Protein (HMGB1) ELISA Kit in biological samples. The concentration gradients of the kit standards or positive controls provide a theoretical detection range of the kit in biological research samples containing HMGB1.

The analytical biochemical ELISA technique of the MBS738320 kit is based on HMGB1 antibody-HMGB1 antigen interactions (immunosorbent) and an HRP colorimetric detection system to detect HMGB1 antigen targets in samples. The ELISA Kit is designed to detect native, non-recombinant HMGB1. Appropriate sample types may include secretions and undiluted body fluids and/or tissue homogenates. Quality control trials evaluating reproducibility identified the intra-assay CV (%) and inter-assay CV (%).

Principle of the assay: The HMGB1 ELISA kit applies the competitive enzyme immunoassay technique using an anti-HMGB1 monoclonal antibody and an HMGB1-HRP conjugate. The test sample and buffer are incubated together with the HMGB1-HRP conjugate on a precoated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for the HRP enzyme.

The enzyme-substrate reaction product forms a blue complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. Color intensity is measured spectrophotometrically at 450 nm in a microplate reader. The intensity of the color is inversely proportional to the HMGB1 concentration since the HMGB1 in the samples and the HMGB1-HRP conjugate compete for the binding site of the anti-HMGB1 antibody.

Since the number of sites is limited, as HMGB1 in the sample occupies more sites, fewer sites remain to bind the HMGB1-HRP conjugate. A standard curve is drawn relating the intensity of the color (O.D.) to the concentration of standards. The HMGB1 concentration in each sample is interpolated from this standard curve.
