Principle of the assay: The HMGB1 ELISA kit applies the competitive enzyme immunoassay technique using an anti-HMGB1 monoclonal antibody and an HMGB1-HRP conjugate. The test sample and buffer are incubated together with the HMGB1-HRP conjugate on a precoated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for the HRP enzyme.
The enzyme-substrate reaction product forms a blue complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. Color intensity is measured spectrophotometrically at 450 nm in a microplate reader. The intensity of the color is inversely proportional to the HMGB1 concentration since the HMGB1 in the samples and the HMGB1-HRP conjugate compete for the binding site of the anti-HMGB1 antibody.
Since the number of sites is limited, as HMGB1 in the sample occupies more sites, fewer sites remain to bind the HMGB1-HRP conjugate. A standard curve is drawn relating the intensity of the color (O.D.) to the concentration of standards. The HMGB1 concentration in each sample is interpolated from this standard curve.
